SUMMARY: Nanoparticle heating has shown promise as a means to cause tumor ablation, improve chemotherapeutic treatment and initiate cancer cell death @ 42-55 C. Targeted temperature responsive magnetic liposomes are used to release a cancer drug directly into tumor tissue.

There is notable synergy when magnetic heating is combined with chemotherapeutic treatment. (1) Weakening the cancer cell’s drug resistance.

(2)       Improved permeability and tumor microvascular pore size to the drug. (3) Efficient and targeted delivery of the cancer drug limits the systemic toxicity.

BACKGROUND ON CANCER TREATMENT: Surgery and Radiation are the 2 most common treatments for cancer. Surgery cannot typically remove all the cancer tissue, especially small metastatic growths. Radiation shrinks cancer tissue. This kills some of the tumor cells. However, the radiation dose is limited by the risk of damaging surrounding tissue. Chemotherapy and other drug treatments are temporarily effective but only until the drug’s effectiveness is reduced by the tumor’s cumulative resistance to the drug and the toxicity of the drug to the healthy cells.

 

Magnetic Hyperthermia is a relatively new concept and cancer treatment. There are two treatment methods being tested. In Vivo injection of nanoparticles into a tumor allows magnetic heating (AMF) to 42-55C tumor ablation, collapse and hemorrhage of the tumor cells. Another method (above) is to use temperature responsive magnetic liposomes to target the tumor. Magnetic heating triggers the release of the chemotherapeutic drug on- command, typically after MRI observation of the concentration of the liposomes in the tumor. Heating further improves tumor blood vessel permeability to the drug and improves the overall efficacy of the treatment.

 

The Magnetic Hyperthermia Instrument has attracted worldwide interest and is primarily used in university and pharmaceutical research laboratories. Small animal testing has so far produced mostly successful results. Recent improvements in the magnetic heating power of the MSI Automation system has resulted in new opportunities for researchers. Virtually all research in the past has been limited to treating mice. Now MSI’s systems are able to provide high magnetic heating power with heating coil diameters up to 120 mm. Other innovations are being considered by MSI Automation to push the magnetic hyperthermia technology to a level where preclinical trials can begin.
